Second gene involved is VKORC1 i.e. vitamin K epoxide reductase complex-1, This enzyme is produced in liver and which is an actual drug target in warfarin treatment. Warfarin acts as an inhibitor for VKORC1 by blocking enzyme vitamin K epoxide reductase. It’s genotypes of this gene has 20-25% variation in drug doses. 1639G>A variant of this gene is highly sensitive to warfarin and since lower dose is required.
USE OF TEST:
Genotyping of these two genes i.e. CYP12C9 and VKORC1 is important before use of warfarin. Known genotypes of these genes helps doctor in decision of precise drug dosing of warfarin. This test has proven its role in timely and precise warfarin use to avoid further severe complications like bleeding.
