Microbial Identification service


16S rRNA Sequencing (full length with phylogeny)

This gold-standard method provides accurate bacterial identification down to the species level by analyzing the highly conserved 16S rRNA gene. We perform full-length sequencing and phylogenetic analysis to determine the evolutionary relationships and nearest matches to known organisms in international databases.


E. coli strain Identity Testing

Specifically for Escherichia coli, we utilize 16S sequence reference-based alignment and
API 20E biochemical profiling for precise identification.

Test of Purity: Bacterial, Fungal & Bacteriophage Screening


Bacterial and fungal impurity

We employ a culture-based approach using four different media and incubation up to 15 days to detect a broad range of potential bacterial and fungal contaminants. We also check for the presence of antibiotics or agents that inhibit the growth of other microbes.


Bacteriophage Detection

We test for the presence or absence of both lytic and lysogenic bacteriophages using positive and negative control cultures to ensure the integrity of your cell bank.

Copy number determination by real-time PCR method

We isolate total DNA and perform quality control. Utilizing primer synthesis for recombinant and reference genes (e.g., dxs in E. coli), we construct standard curves and assess linearity and extraction efficiency. SYBR Green Real-time qPCR is used to estimate the absolute transgene copy number. Testing is performed on three vials.



Restriction endonuclease analysis

This technique helps to verify the integrity and expected structure of the genetic material in the cell bank.



Retention of selectable marker

We determine plasmid stability by assessing the retention of the selectable marker on both selective and non-selective media.


Retention of recombinant construct

We evaluate the stability of the recombinant construct over three subcultures, followed by DNA sequencing of the Open Reading Frame (ORF) to ensure sequence integrity.

Why is 16S rRNA sequencing important for bacterial cell bank identity testing?

16S rRNA gene sequencing is a widely used molecular method for confirming bacterial identity because the gene contains conserved regions for broad bacterial detection along with variable regions that support taxonomic identification. Full-length 16S rRNA sequencing combined with phylogenetic analysis provides a robust assessment of the organism’s relationship to established reference strains.

How do you test microbial cell banks for contamination and bacteriophages?

We perform culture-based microbial purity testing using multiple media and extended incubation to detect bacterial and fungal contaminants, including organisms that may grow under different culture conditions. We also assess whether antibiotics or other inhibitory agents could mask contamination by suppressing microbial growth.

Bacteriophage testing evaluates the presence or absence of bacterial viruses that can compromise bacterial cell banks, fermentation performance, and manufacturing consistency. Cell-bank testing programs commonly include assessment for lytic, lysogenic, and/or prophage-related risks, particularly for E. coli and other bacterial production hosts.

Do you characterize both Master Cell Banks (MCB) and Working Cell Banks (WCB)?

Yes. Characterization is performed at multiple points in the cell banking lifecycle: parental/pre-MCB material, MCB, WCB, and End-of-Production (EOP) cells.  MCBs typically receive full characterization, while WCBs undergo an abbreviated but compliant set of tests, with additional assessments at the limit of in vitro cell age as required by ICH Q5A(R1)/Q5D.

What samples are required for cell bank characterization, and how long does testing take?

We generally require frozen vials from your Master Cell Bank (MCB)Working Cell Bank (WCB), or research cell bank, together with relevant strain history, host information, plasmid/construct maps, sequence references, growth conditions, and applicable acceptance criteria. The exact number of vials required depends on the selected test panel, organism, and whether confirmatory or repeat testing may be needed.

Turnaround time depends on the scope of testing. Identity or viability testing may be completed sooner, whereas extended microbial purity testing, bacteriophage detection, subculture-based genetic stability studies, and sequencing require additional time. We recommend defining the complete cell bank characterization plan early so testing can be aligned with your research, manufacturing, or regulatory timeline.
